Journal of pharmaceutical and biomedical analysis, Volume 6, Issue 6-8, 1 1 1988, Pages 895-902 Evaluation of sample work-up methods and internal standards for the determination of catecholamines in urine by HPLC with electrochemical detection. Claeys M, Schepers A, Dillen L, De Potter WP
Two sample work-up methods: (I) one consisting of adsorption of the catecholamines onto alumina followed by ion pair extraction and (II) another consisting of isolation by cation exchange and subsequent adsorption onto alumina, have been evaluated for the assay of urinary catecholamines by means of HPLC with electrochemical detection. With the aim of achieving high precision, two internal standards, i.e. dihydroxybenzylamine and epinine, have been compared. The results indicate that clean HPLC c... Abstract

Nature, Volume 330, Issue 6148, 1 1 1987, Pages 578-581 Deletion of a DNA sequence at the chromosomal region 3p21 in all major types of lung cancer. Kok K, Osinga J, Carritt B, Davis MB, van der Hout AH, van der Veen AY, Landsvater RM, de Leij LF, Berendsen HH, Postmus PE
In childhood malignancies such as retinoblastoma and Wilms tumour, of which both familial and sporadic forms exist, recessive mutations of presumed differentiation genes have been implicated in tumorigenesis. A proportion of cases appear with microscopically visible chromosome deletions which indicate the regions where the genes concerned are located. Mutation or loss of one allele causes a cancer predisposition. For tumour development functional loss of the remaining normal allele is also requi... Abstract

Molecular immunology, Volume 24, Issue 10, 1 1 1987, Pages 1047-1053 Complement-mediated solubilization of rat IgA immune precipitates. Rits M, Hiemstra PS, van Es LA, Bazin M, Vaerman JP, Daha MR
The complement-mediated solubilization (CMS) of immunoprecipitates (IP) consisting of DNP-rat serum albumin (RSA) and rat monoclonal anti-DNP antibodies of the IgA [both polymeric (p-) and monomeric (m-)] or IgG2b (sub)class was studied. In contrast to IgG2b IP, solubilization of IgA IP was only observed in an autologous system, with rat serum as the source of complement. IP prepared using m-IgA were solubilized faster than those prepared using p-IgA. Analysis of both affinity and avidity of the... Abstract

European journal of cancer & clinical oncology, Volume 23, Issue 9, 1 1 1987, Pages 1409-1411 Retreatment with the induction regimen in small cell lung cancer relapsing after an initial response to short term chemotherapy. Postmus PE, Berendsen HH, van Zandwijk N, Splinter TA, Burghouts JT, Bakker W
In 37 patients with small cell lung cancer treatment with five cycles of cyclophosphamide, doxorubicin and etoposide (CDE), resulted in 23 complete (CR) and 14 partial responses (PR). Median response duration was 34 weeks. At relapse all patients were retreated with CDE. In 23 (62%) patients this gave a second response (6 CR, 17 PR). Factors influencing the occurrence of a second response were: 1. a CR after the first five cycles of CDE; 18 out of 23 CR patients responded again whereas only five... Abstract

Cancer genetics and cytogenetics, Volume 27, Issue 2, 1 1 1987, Pages 361-365 Loss of heterozygosity for a chromosome 3 sequence presumably at 3p21 in small cell lung cancer. Mooibroek H, Osinga J, Postmus PE, Carritt B, Buys CH
A recombinant DNA fragment detecting a chromosome #3 restriction fragment length polymorphism presumably at p21 was hybridized to HindIII-digested DNA isolated from the leukocytes of 12 patients of small cell lung cancer. Four of them appeared to be heterozygous. Analysis of tumor material from these four patients revealed homozygosity for either one or the other restriction fragment in every case. Our findings suggest the presence on the short arm of chromosome #3 of a recessive mutant cancer g... Abstract

Scandinavian journal of immunology, Volume 26, Issue 2, 1 1 1987, Pages 111-117 The IgA-binding lectin jacalin induces complement activation by inhibition of C-1-inactivator function. Hiemstra PS, Gorter A, Stuurman ME, Van Es LA, Daha MR
Jacalin, a D-galactose-specific lectin from jackfruit, interacts with human IgA and one or two other serum proteins. Incubation of jacalin with fresh human serum was shown to result in activation of the complement system. Therefore the mechanism of complement activation by jacalin was studied. Jacalin was extracted from jackfruit seeds (crude preparation) and purified to homogeneity by affinity chromatography on IgA-Sepharose to yield a pure preparation of jacalin. Both crude and pure jacalin we... Abstract

European journal of cancer & clinical oncology, Volume 23, Issue 8, 1 1 1987, Pages 1207-1208 Phase II evaluation of trans-N3P3Az2(NHMe)4 (AZP) in non-small cell lung cancer. Postmus PE, Mulder NH, Grampel JC, Meyers WH, Berendsen HH
Trans-N3P3Az2(NHMe)4 (AZP), an aziridinyl substituted cyclophosphazene, was evaluated in a phase II study in non-small cell lung cancer at a dose of .33 mg/m2 i.v. bolus every 3 weeks. There were no tumor responses seen. Cumulative bone marrow toxicity, comparable to other cyclophosphazenes, was noted. AZP is not useful for the treatment of NSCLC. Abstract

Immunology, Volume 61, Issue 3, 1 1 1987, Pages 303-309 IgA- and secretory IgA-opsonized S. aureus induce a respiratory burst and phagocytosis by polymorphonuclear leucocytes. Gorter A, Hiemstra PS, Leijh PC, van der Sluys ME, van den Barselaar MT, van Es LA, Daha MR
The aim of the present study was to investigate whether corpuscular immune complexes containing human IgA were able to interact with human polymorphonuclear leucocytes (PMN). As a model for corpuscular IgA immune complexes (IgA IC), heat-killed Staphylococcus aureus (S. aureus) opsonized with either purified human serum IgA or purified secretory IgA (sIgA) isolated from human colostrum was used. In order to determine the capacity of IgA and sIgA to opsonize S. aureus the phagocytosis of these Ig... Abstract

Research communications in chemical pathology and pharmacology, Volume 57, Issue 1, 1 1 1987, Pages 15-32 Quantification of myocardial necrosis and cardiac hypertrophy in isoproterenol-treated rats. Knufman NM, van der Laarse A, Vliegen HW, Brinkman CJ
Administration of a single high dose or multiple low doses of isoproterenol (ISO) to rats induces myocardial necrosis and cardiac hypertrophy. To assess the extent of necrosis we measured myocardial activities of creatine kinase (CK) and aspartate aminotransferase (AST) in the first days after ISO-treatment. After a single high dose and multiple low doses of ISO, necrosis was 10% and 25%, respectively. To quantify the degree of hypertrophy we measured heart weight and myocyte volume, and calcula... Cardiovascular research, Volume 21, Issue 5, 1 1 1987, Pages 352-357 Morphometric quantification of myocyte dimensions validated in normal growing rat hearts and applied to hypertrophic human hearts. Vliegen HW, van der Laarse A, Huysman JA, Wijnvoord EC, Mentar M, Cornelisse CJ, Eulderink F
To obtain quantitative information on changes in myocyte size in hearts with increased myocardial mass light microscopical morphometry was used in fixed and stained myocardial tissue sections. A modification of the azan staining procedure made longitudinal (sarcolemma) and transverse (intercalated discs) boundaries visible with a contrast that allowed morphometric analysis. The method was validated in left ventricular tissue of rats ranging in age from 4 to 17 weeks. Myocyte width, cut surface a... Abstract
