American journal of respiratory and critical care medicine, Volume 156, Issue 6, 1 1 1997, Pages 1765-1772 Effects of photochemical air pollution and allergen exposure on upper respiratory tract inflammation in asthmatics. Hiltermann TJ, de Bruijne CR, Stolk J, Zwinderman AH, Spieksma FT, Roemer W, Steerenberg PA, Fischer PH, van Bree L, Hiemstra PS
Asthma is an inflammatory disease of the airways, and exacerbations of this disease have been associated with high levels of air pollution. The objective of this study was to examine whether ambient air pollution and/or allergen exposure induces inflammatory changes in the upper airways of asthmatics. Sixty patients with intermittent to severe persistent asthma visited the Hospital's Out Patient Clinic every 2 wk for a period of 3 mo, and on each visit a nasal lavage was obtained. Associati... Revue internationale de recherches de readaptation, Volume 20, Issue 3, 1 1 1997, Pages 289-301 The quality of life of people with mental retardation: in search of an adequate approach. Vreeke GJ, Janssen CG, Resnick S, Stolk J
There is a lack of instruments that measure the quality of life of people with mental retardation. These types of instruments could be used in order to give an indication of the quality of care they receive. At the moment we are developing an instrument that measures quality of life. Our first task is to find an adequate definition of 'quality of life'. In this article an attempt is made to define this term as it relates to people with mental retardation. Starting from literature in th... Abstract

Current drug treatment of pulmonary emphysema has little effect on quality of life and duration of survival. Surgical treatment for patients with severe pulmonary emphysema was recently introduced; it consists of resection of lung tissue with poor ventilation and perfusion. Surgical reduction of lung volume improves the forced expiratory pressure per second by 80-100% from 0.8 to 1.51. This increases the exercise tolerance and improves the patient's functioning in everyday life. The postope... Abstract

European journal of clinical investigation, Volume 27, Issue 2, 1 1 1997, Pages 148-156 An enzyme immunoassay for polymorphonuclear leucocyte-mediated fibrinogenolysis. Bos R, van Leuven CJ, Stolk J, Hiemstra PS, Ronday HK, Nieuwenhuizen W
Upon stimulation, polymorphonuclear leucocytes (PMNs) release potent serine proteases, i.e. elastase, cathepsin G and proteinase 3, which contribute to the degradation of tissue and plasma components. Here, we describe the development of a plasma test to assess PMN-mediated fibrinogenolysis as a biochemical marker for actual PMN-derived proteolysis in vivo, useful for monitoring therapeutic efficacy, i.e. of elastase inhibitors. We generated a monoclonal antibody (MAb), designated 1-1/B3, with a... Abstract

Antileukoprotease (ALP), or secretory leukocyte proteinase inhibitor, is an endogenous inhibitor of serine proteinases that is present in various external secretions. ALP, one of the major inhibitors of serine proteinases present in the human lung, is a potent reversible inhibitor of elastase and, to a lesser extent, of cathepsin G. In equine neutrophils, an antimicrobial polypeptide that has some of the characteristics of ALP has been identified (M. A. Couto, S. S. L. Harwig, J. S. Cullor, J. P... Abstract

The Journal of experimental medicine, Volume 184, Issue 4, 1 1 1996, Pages 1305-1312 Colony growth of human hematopoietic progenitor cells in the absence of serum is supported by a proteinase inhibitor identified as antileukoproteinase. Goselink HM, van Damme J, Hiemstra PS, Wuyts A, Stolk J, Fibbe WE, Willemze R, Falkenburg JH
Serum contains many growth factors and nutrients that stimulate colony formation of hematopoietic progenitor cells (HPC) in semisolid cultures. In the absence of serum, no proliferation of HPCs could be obtained in semisolid medium cultures of partially purified bone marrow cells in the presence of multiple hematopoietic growth factors, insulin, cholesterol, and purified clinical-grade human albumin. This appeared to be due to a suppressive activity induced by monocyte- and T lymphocyte-depleted... Abstract

The Netherlands journal of medicine, Volume 48, Issue 1, 1 1 1996, Pages 8-10 Complications in the use of the subcutaneous tunnelled intratracheal oxygen catheter. in't Veen JC, Stolk J, Dijkman JH
Transtracheal oxygen delivery seems to be a safe procedure in the treatment of chronic obstructive pulmonary disease (COPD) with chronic hypoxaemia. Even so, serious complications do occur. Three patients in whom we used a subcutaneous tunnelled intratracheal oxygen catheter (ITO2C) are described. Surgical intervention was required in all because of complications from the procedure. One of the complications--tracheal and catheter obstruction with stridor and subcutaneous emphysema by granulomato... Abstract

Clinical science (London, England : 1979), Volume 89, Issue 6, 1 1 1995, Pages 619-624 Effect of ozone exposure on maximal airway narrowing in non-asthmatic and asthmatic subjects. Hiltermann TJ, Stolk J, Hiemstra PS, Fokkens PH, Rombout PJ, Sont JK, Sterk PJ, Dijkman JH
1. Ozone is a major constituent of air pollution in the summer. Epidemiological studies have demonstrated that there is an increase in hospital admissions for respiratory diseases 1 day after peak levels of ambient ozone. This may be due to an increase in the responsiveness of the airways to bronchoconstrictor stimuli. 2. In the present study we therefore studied the effect of a controlled exposure to ozone on the maximal degree of airway narrowing to a non-specific bronchoconstrictor, methachol... Abstract

Background: The neutrophil elastase inhibitor, secretory leucocyte protease inhibitor (SLPI), is a potential therapeutic tool in inflammatory lung diseases such as cystic fibrosis and pulmonary emphysema. The distribution and disappearance in the lung of aerosolised recombinant SLPI (rSLPI) was investigated in healthy humans and in patients with cystic fibrosis or alpha 1-antitrypsin-associated emphysema. Methods: To distinguish aerosolised rSLPI from endogenous SLPI the recombinant inhibitor wa... Abstract

Blood coagulation & fibrinolysis : an international journal in haemostasis and thrombosis, Volume 6, Issue 3, 1 1 1995, Pages 259-267 A monoclonal antibody with high affinity for a neo-antigenic site in fibrinogen degraded by polymorphonuclear leukocyte-derived elastase. Bos R, van Leuven CJ, Stolk J, Hiemstra PS, Dijkman JH, Nieuwenhuizen W
Elastase, released by stimulated polymorphonuclear leukocytes (PMN), is thought to play an important role in the pathogenesis of chronic obstructive pulmonary disease (COPD) especially pulmonary emphysema. A test that can detect release of elastase activity from PMN would be valuable to monitor therapy or to identify patients at risk. The authors aimed to isolate and characterize monoclonal antibodies (mAb) with a high affinity for a neo-antigenic determinant in a high-molecular weight degradati... Abstract
